چکیده
Aquatic environmental DNA (eDNA) studies have considerably taken off during the last 10 years, promptly establishing eDNA as a new approach to monitoring ecological status of European surface waters. On one hand due numerous biological methods based on aquatic species, other development group-specific primers that allowed discrimination between species or at least genera. Understandably, rapid achievements were made for animal groups include small number (fish, amphibians) whereas it is still in plants, complexity obtaining universal barcode applicable latter. Nonetheless, research plant barcoding and metabarcoding making significant progress, mainly expanding detection freshwater ecosystems. With multi-barcode mind, we are preparing reference library our study area, Northern Vosges Regional Natural Park. Drawing from this experience supported by current publications, focus strong connection floristic inventories, local libraries, herbaria. Prior knowledge distribution abundant genetic data key increasing through (Cordier et al. 2020). We ought standardise practice before plants can cover large river basins span time. Following consensus combination barcodes kingdom, most recurrent ones plastid trn L, mat K, rbc L regions nrDNA internal transcribed spacers (ITS) although likely emerge further research. To date, selection seems be utmost importance since they offer different properties question. Seminal efforts focused species-specific assays tailor-made primers. These demonstrated reliability used species. Today, three common invaders, Elodea canadensis , nutalli Hydrilla verticillata now their own aiming identify earliest invasions (Gantz 2018). contrary, “universal” primers, such those White (1990) within ITS, prove useful where many differing families involved (Coghlan Despite fast-moving eDNA, libraries lack hindering progress applying surveys metabarcoding. Such rely heavily building which extremely time consuming require important taxonomic encumbered polyphyly groups.

Potential for DNA-based identification of Great Lakes fauna: match and mismatch between taxa inventories and DNA barcode libraries
DNA-based identification of mixed-organism samples offers the potential to greatly reduce the need for resource-intensive morphological identification, which would be of value both to bioassessment and non-native species monitoring. The ability to assign species identities to DNA sequences found depends on the availability of comprehensive DNA reference libraries. Here, we compile inventories f...
متن کاملImproved protocols to accelerate the assembly of DNA barcode reference libraries for freshwater zooplankton
Currently, freshwater zooplankton sampling and identification methodologies have remained virtually unchanged since they were first established in the beginning of the XX century. One major contributing factor to this slow progress is the limited success of modern genetic methodologies, such as DNA barcoding, in several of the main groups. This study demonstrates improved protocols which enable...
متن کاملA DNA barcode for land plants.
DNA barcoding involves sequencing a standard region of DNA as a tool for species identification. However, there has been no agreement on which region(s) should be used for barcoding land plants. To provide a community recommendation on a standard plant barcode, we have compared the performance of 7 leading candidate plastid DNA regions (atpF-atpH spacer, matK gene, rbcL gene, rpoB gene, rpoC1 g...
متن کاملRefining the DNA barcode for land plants.
T he goal of DNA barcoding is conceptually simple: Find one or a few regions of DNA that will distinguish among the majority of the world’s species, and sequence these from diverse sample sets to produce a large-scale reference library of life on earth (1). This approach can then be used as a tool for species identification and to help in the discovery of new species (2). Since the first DNA ba...
متن کاملUsing herbarium-derived DNAs to assemble a large-scale DNA barcode library for the vascular plants of Canada1
Premise of the study Constructing complete, accurate plant DNA barcode reference libraries can be logistically challenging for large-scale floras. Here we demonstrate the promise and challenges of using herbarium collections for building a DNA barcode reference library for the vascular plant flora of Canada. Methods Our study examined 20,816 specimens representing 5076 of 5190 vascular plant ...
